Background

Lysosomal associated membrane protein 2 (LAMP2) is a heavily glycosylated single-pass integral membrane protein encoded by the LAMP2 gene, enriched on late endosomal and lysosomal limiting membranes with multiple tissue-specific splicing isoforms. LAMP2 carries dense luminal glycan chains that form a protective barrier against luminal acid hydrolases, and interacts with endocytic adaptor proteins to mediate autophagosome-lysosome fusion and endocytic cargo sorting. Loss of intact LAMP2 glycoprotein scaffold will trigger lysosomal membrane leakage and massive accumulation of undegraded intracellular waste substrates.

LAMP2 exerts dual homeostatic functions via two core structural domains: the heavily glycosylated extracellular region shields organelle lipid bilayers from hydrolytic degradation, while the short cytoplasmic tail binds vesicle trafficking adaptors to guide autophagic and endocytic cargo into lysosomal lumens. Unlike LAMP1 with partial functional redundancy, LAMP2 is irreplaceable for autophagosome tethering; LAMP2 deletion blocks autophagy flux completely, uncoupling cellular waste clearance from lysosomal degradative capacity. This dual regulatory characteristic balances steady-state cellular turnover under physiological conditions, while LAMP2 loss-of-function mutations lead to severe multi-organ lysosomal storage lesions. LAMP2 participates in broad biological processes including autophagy circulation, lysosomal morphological maturation and inherited storage diseases.
