Membrane frizzled-related protein (MFRP) is a single-pass membrane-associated protein encoded by the MFRP gene, anchored to plasma membrane outer surfaces within ocular somatic cell populations. Unlike soluble cytoplasmic regulatory proteins lacking lipid-binding helices, MFRP carries one transmembrane segment and conserved extracellular cysteine-rich frizzled domains for secreted morphogen ligand recognition. It appears to function as a membrane-bound modulator to buffer extracellular morphogen signal levels under physiological tissue development conditions.
MFRP is a type II single-pass membrane protein enriched at the apical membrane of retinal pigment epithelium and ciliary epithelium cells, where it participates in membrane organization and protein interaction networks. Its extracellular region contains CUB, LDLa, and Frizzled-related domains that contribute to its structural and interaction properties. Rather than acting as a general morphogen-sequestering or signal-buffering receptor, MFRP functions as a membrane-associated molecular hub whose biological roles appear to depend on interactions with specific membrane proteins and lipids. Reported interaction partners include C1QTNF5, ADIPOR1, and KCNJ13, linking MFRP to membrane organization, protein localization, and lipid homeostasis in ocular tissues. These interactions may help coordinate the organization and functional stability of specialized membrane domains, although the precise molecular mechanisms of MFRP remain under investigation.
MFRP may modulate intracellular developmental signaling cascades triggered by extracellular morphogen ligand sequestration. Sequence variants within the MFRP gene alter morphogen ligand binding affinity and correlate with disorganized tissue structural states. No other membrane frizzled-related protein fully recapitulates MFRP’s dual capacity for secreted morphogen sensing and membrane-localized signal buffering at plasma membranes. Variations in MFRP expression levels likely correspond to tissue developmental patterning status, rendering the protein a suitable research target for frizzled domain structural biology and morphogen gradient signal analysis.
MFRP localizes to plasma membrane outer leaflets to bind secreted extracellular morphogens without entering cytoplasmic organelle lumens. Its integral membrane topology separates it from soluble cytoplasmic regulatory factors, with dual potential roles in sustaining steady tissue morphogen gradients and mediating ordered tissue structural patterning cues. MFRP-dependent morphogen buffering may adjust intracellular developmental messenger concentrations and modify cellular tissue growth activity. Reduced functional MFRP elevates unregulated extracellular morphogen signal levels and weakens secreted ligand response capacity, making MFRP a research subject for basic membrane frizzled-related protein studies.

The biological functions of integral membrane MFRP frizzled-related protein are focused on sustained extracellular morphogen binding and cytoplasmic developmental signal cascade transmission:
Creative Biolabs offers purified MFRP membrane protein samples produced under unified preparation workflows, including full-length MFRP constructs and isolated extracellular frizzled ligand-binding domain variants. Truncated protein fragments cannot support complete morphogen ligand sequestration and developmental signal buffering activity, while full-length constructs fit research focused on membrane frizzled-morphogen interaction and extracellular gradient conformational analysis. All batches receive uniform quality screening. Functional assessments may only be performed on full-length membrane protein under simulated lipid bilayer microenvironment setups. Consistent extracellular frizzled domain structural features are preserved across batches to support comparative laboratory analysis between experimental groups. Full-length MFRP membrane samples retain intact cysteine-rich morphogen ligand-binding fold conformation after standardized purification, which supports reliable detection of weak and transient morphogen-protein binding events in comparative membrane functional analysis.
