Magnesium transporter MRS2 (MRS2) is a mitochondrial inner-membrane cation channel encoded by the MRS2 gene. Each MRS2 protomer contains two transmembrane helices and a large matrix-facing N-terminal domain, and five protomers assemble into a homopentameric pore. Structural and electrophysiological studies support a major role for MRS2 in mitochondrial Mg2+ uptake and homeostasis, while permeability to other cations has also been reported under experimental conditions. Unbalanced mitochondrial magnesium concentrations can disrupt Mg2+-dependent enzymatic activity, and MRS2 contributes to cation flux between the cytosol and mitochondrial matrix. Functional MRS2 assemblies help maintain mitochondrial magnesium homeostasis by regulating cation entry into the matrix. Conserved pore residues contribute to ion coordination and channel regulation rather than establishing absolute Mg2+ selectivity.
Sequence variants within MRS2 coding regions might alter pore permeability and shift mitochondrial magnesium homeostasis. Related cation transport proteins may share partial functional features, but MRS2 has distinct channel and regulatory properties in the mitochondrial inner membrane. MRS2 is embedded in the mitochondrial inner membrane and does not exist as a freely soluble cytoplasmic protein. Its two-transmembrane topology and pentameric assembly distinguish it from soluble metal-binding factors and support cation uptake into the mitochondrial matrix. Diminished functional MRS2 can disrupt mitochondrial enzyme activity reliant on magnesium cofactors, rendering this channel a suitable target for mitochondrial cation transport research.

The biological functions of mitochondrial inner-membrane MRS2 channel are focused on cation permeation and regulation of mitochondrial magnesium homeostasis:
Creative Biolabs offers purified MRS2 membrane protein samples produced under unified preparation workflows, including full-length MRS2 constructs and isolated pore domain variants. Truncated polypeptide fragments cannot support complete channel activity, while full-length constructs fit mitochondrial cation transport research. All batches receive uniform quality screening. Functional assessments may only be performed under simulated lipid bilayer microenvironment setups. Transmembrane pore structural features are preserved across batches to support comparative laboratory analysis between experimental groups. Full-length MRS2 membrane samples retain intact cation coordination residues after standardized purification, which supports reliable detection of weak and transient ion-pore binding events in comparative membrane functional analysis.

Creative Biolabs provides cell research models with adjustable MRS2 expression levels, suitable for structural observation of mitochondrial inner-membrane cation channels and magnesium homeostasis research. Sample assessment covers sustained mitochondrial membrane localization detection and preliminary cation transport analysis, enabling side-by-side comparison of transport behavior under varying membrane expression abundances. These cell models can be paired with cation quantification schemes to track mitochondrial magnesium shifts linked to MRS2 dosage changes.
