Background

Mucin 4 (MUC4) is a single‑pass transmembrane mucin molecule encoded by MUC4 gene. This protein localizes predominantly on the apical plasma membrane of epithelial cell populations across multiple mucosal tissues. MUC4 contains large extracellular glycosylated domains exposed toward extracellular space alongside a short intracellular cytoplasmic segment. Unlike many membrane receptors that trigger robust downstream signalling upon ligand engagement, the heavily glycosylated extracellular region forms part of the epithelial surface coating layer, shaping the physicochemical properties of cell surface interface.

Mucin‑rich surface layers constitute a critical interface between host tissue and surrounding extracellular milieu. Variations in MUC4 surface abundance could alter the profile of surface‑presented glycan moieties, which might influence molecular recognition events between adjacent cells and surrounding soluble factors. Altered MUC4 expression appears to correlate with shifts in tissue surface characteristics, rendering this molecule a meaningful research target for epithelial barrier biology and cell‑surface interaction studies. Other mucin family members cannot fully recapitulate the distinct structural features contributed by MUC4 within mucosal tissue compartments. Changes to MUC4 surface presentation may contribute to modified cell‑to‑cell recognition profiles within tissue microenvironments.

MUC4 Protein Function: Core Roles in Epithelial Surface Coating and Intercellular Molecular Recognition

The biological functions of MUC4 are focused on epithelial surface barrier formation, glycan‑dependent molecular recognition and tissue microenvironment modulation:

  • Extensive Glycosylated Extracellular Scaffold: Carries heavily modified extracellular repeat domains that shape the physicochemical features of epithelial cell surface. Might participate in non-covalent intermolecular contacts with diverse surrounding soluble and cell-surface partners.
  • Epithelial Surface Modulation: Contributes to assembling the outer surface coating of mucosal epithelia, and could adjust surface accessibility for surrounding molecular components.
  • Intercellular Recognition Mediator: Is proposed to support molecular contacts between neighbouring cell populations via surface glycan structures.
  • Tissue Compartment Adaptation: Variable expression levels across mucosal sites may correspond to local demands for surface barrier properties.
  • Research Model Relevance: Modified MUC4 structural features might reshape cell-surface interaction profiles within research observation systems.

MUC4 Membrane Protein Product

Creative Biolabs offers purified MUC4 membrane samples via standardized preparation workflows, including full‑length MUC4 constructs and isolated extracellular structural variants. Isolated extracellular fragments may not support complete intermolecular interaction related behaviours, while full‑length constructs may be suited for cell‑surface molecular recognition and mucosal barrier related research. All samples receive routine quality screening, and functional relevant observation may only be carried out with full‑length samples under simulated membrane environments. All sample batches follow unified processing standards to maintain consistent structural features for comparative laboratory analysis across separate test groups.
