Background

The PDGFRA gene at 4q12 encodes platelet-derived growth factor receptor alpha, a type I transmembrane glycoprotein of 1,089 amino acids belonging to the class III receptor tyrosine kinase family. PDGFRA contains five extracellular immunoglobulin-like domains, a single transmembrane helix, and a split intracellular tyrosine kinase domain with a kinase insert. The mature glycosylated protein is approximately 170 kDa and is expressed on mesenchymal cells, oligodendrocyte precursor cells, fibroblasts, and vascular smooth muscle cells. PDGFRA is activated by PDGF-AA, PDGF-BB, and PDGF-CC homodimers, which induce receptor dimerization, autophosphorylation, and activation of PI3K/AKT, MAPK/ERK, PLCγ, and STAT pathways driving cell proliferation, migration, and survival. PDGFRA signaling is essential for embryonic development including neural crest migration and organogenesis. Activating PDGFRA mutations drive gastrointestinal stromal tumors, with the D842V substitution being the most common imatinib-resistant variant. PDGFRA amplifications occur in glioblastoma, while chromosomal rearrangements such as FIP1L1-PDGFRA are found in specific myeloid and lymphoid neoplasms. PDGFRA is therefore an important target in oncology, developmental biology, and receptor tyrosine kinase research, providing tools for understanding growth factor receptor signaling and kinase-targeted therapeutics.

Function of PDGFRA Protein: A Receptor Tyrosine Kinase in Cell Growth and Oncogenic Signaling

The functional spectrum of PDGFRA spans developmental signaling and oncogenic biology:

  • Ligand-dependent activation: PDGFRA binds PDGF-AA, PDGF-BB, and PDGF-CC, inducing dimerization and trans-autophosphorylation. Phosphotyrosine sites recruit PI3K, PLCγ, and Grb2-SOS, activating MAPK/ERK and PI3K/AKT pathways that promote cell proliferation, survival, and migration.
  • Developmental roles: PDGFRA signaling drives neural crest migration, craniofacial morphogenesis, lung alveolarization, and oligodendrocyte development. In adults, PDGFRA maintains mesenchymal stem cells and regulates wound healing through fibroblast recruitment.
  • Oncogenic activation: Activating PDGFRA mutations occur in 5-8% of GIST, with D842V conferring imatinib resistance. PDGFRA amplification at 4q12 occurs in glioblastoma, and PDGFRA fusions drive dermatofibrosarcoma and hypereosinophilic syndromes.
  • Therapeutic targeting: PDGFRA is inhibited by imatinib, sunitinib, and the D842V-selective agent avapritinib, which is approved for PDGFRA D842V-mutant GIST. PDGFRA inhibitors are also under investigation in glioblastoma and other PDGFRA-driven malignancies.

PDGFRA Stable Cell Line Product

Constructing a robust cell model is essential for investigating PDGFRA-mediated signaling and oncogenic transformation. As a receptor tyrosine kinase that drives proliferation through PI3K/AKT and MAPK pathways, PDGFRA expression level and kinase activity directly determine downstream signaling and cellular responses. Activating mutations may lead to constitutive kinase activity and oncogenic transformation. Creative Biolabs offers customized PDGFRA membrane protein stable transfection cell models, encompassing wild-type and oncogenic variant forms including D842V. These models express the target protein on the plasma membrane, and are suitable for studies on PDGFRA kinase activation, ligand-stimulated signaling, evaluation of variant-dependent kinase activity, and drug response profiling, providing a reliable platform for exploring PDGFRA structure-function relationships, oncogenic mutations, and kinase-targeted therapeutic development. Our technology ensures cell membrane integrity and correct protein localization, facilitating your research.

Frequently Asked Questions (FAQ)

  1. What is the main function of the PDGFRA gene?

PDGFRA encodes platelet-derived growth factor receptor alpha, a class III receptor tyrosine kinase activated by PDGF-AA, PDGF-BB, and PDGF-CC. Ligand binding induces receptor dimerization and autophosphorylation, activating PI3K/AKT, MAPK/ERK, and PLCγ signaling pathways that regulate cell proliferation, migration, and survival. PDGFRA is essential for embryonic development including neural crest migration and organogenesis.

  2. Why is PDGFRA an important research target?

Activating PDGFRA mutations drive gastrointestinal stromal tumors, with the D842V variant conferring imatinib resistance and targeted by the selective inhibitor avapritinib. PDGFRA amplification occurs in glioblastoma, and chromosomal translocations generate oncogenic PDGFRA fusions in dermatofibrosarcoma protuberans. PDGFRA is therefore a clinically validated target in precision oncology and kinase biology.
