Background

TNF receptor superfamily member 13B (TNFRSF13B, also known as TACI) is a type III transmembrane receptor encoded by the TNFRSF13B gene, selectively expressed on mature B cells and plasma cells. Its extracellular cysteine-rich tandem motifs bind trimeric BAFF and APRIL; ligand crosslinking triggers receptor oligomerization and activates canonical & non-canonical NF-κB signaling via cytoplasmic adaptors.

Under steady immune conditions, moderate TACI signaling maintains peripheral B cell pools, mediates Ig class switching and sustains long-lived plasma cell antibody secretion. Upon pathogen stimulation, elevated BAFF/APRIL amplifies this pathway to expand antigen-specific B clones. Loss-of-function TNFRSF13B mutations weaken antibody production and induce common variable immunodeficiency, while overactive signaling accumulates autoreactive B cells and drives autoimmune inflammation. As illustrated in the schematic, germline TACI mutation impairs B cell activation and plasma differentiation while elevating early self-reactivity, making TNFRSF13B a core research target for B cell biology and humoral immune disorder mechanism research.
