TNF superfamily member 14 (TNFSF14, LIGHT) is a dual membrane-bound and secreted TNF ligand encoded by TNFSF14 gene, expressed in T lymphocytes, dendritic cells and stromal subsets, localized to plasma membrane and interstitial fluid compartments. Distinct from other TNF cytokines, TNFSF14 engages two distinct TNF receptors to coordinate endothelial upregulation of adhesion molecules and drive lymphoid tissue organogenesis. Under steady immune conditions, basal TNFSF14 expression maintains mild vascular permissiveness without excessive leukocyte infiltration. Upon inflammatory or antigenic stimulation, accumulated ligand triggers endothelial signaling to boost leukocyte recruitment and support germinal center development. Unlike general inflammatory TNF factors, TNFSF14 carries lymphoid-organizing functions that cannot be fully substituted by other superfamily members, linking lymphocyte-stroma crosstalk to secondary lymphoid tissue assembly pathways. Loss of functional TNFSF14 impairs lymphoid structure formation and dampens regional immune responses, while sustained ligand expression amplifies vascular inflammatory recruitment, establishing TNFSF14 as a core research target for lymphoid TNF ligand biology and autoimmune modulator screening.
TNFSF14 executes homotrimeric ligand function embedded in lymphocyte membranes and extracellular space, utilizing conserved TNF fold motifs to selectively bind HVEM and LTβR receptor ectodomains. Conserved trimerization interfaces separate its receptor selectivity from canonical inflammatory TNF paralogs. TNFSF14-mediated receptor clustering bridges lymphocyte-stromal contact and endothelial transcriptional reprogramming, balancing lymphoid organ growth and tissue inflammatory magnitude according to antigen exposure load. TNFSF14 participates in lymph node genesis, mucosal immune cell recruitment and chronic autoimmune inflammation. Deficient TNFSF14 expression disrupts secondary lymphoid tissue architecture and weakens adaptive immune surveillance. Therefore, TNFSF represents a pivotal research target for lymphotropic TNF ligand study and anti-autoimmune compound screening.
