Background

Tyrosinase-related protein 1 (TYRP1) is a type I melanosomal transmembrane glycoprotein encoded by the TYRP1 gene, selectively expressed in melanocytes and melanoma cells and localized to mature pigment organelles, where it participates in melanosome function and melanogenesis. Although DHICA oxidase activity has been reported for Tyrp1 in mouse experimental systems, recombinant human TYRP1 has not demonstrated an established DHICA oxidase activity. TYRP1 has also been implicated in the regulation and stabilization of tyrosinase, thereby supporting normal melanosomal function and pigment production.

Human TYRP1 contains a binuclear zinc-binding site rather than a conserved copper-containing catalytic pocket, and its precise catalytic function remains under investigation. TYRP1 may influence melanogenesis through its roles in melanosome organization and interactions with tyrosinase. TYRP1 variants can disrupt melanosome function and pigmentation, but they should not be assumed to cause DHICA quinone accumulation, oxidative stress or apoptotic death through a defined catalytic mechanism. TYRP1 participates in epidermal pigment formation and has been widely investigated in melanocyte biology and cutaneous melanoma, making it an important research marker for melanogenesis and melanoma-related studies.
