Background

U22 glycoprotein is a betaherpesvirus-encoded integral membrane glycoprotein encoded by viral U22 open reading frame, expressed on infected host cell and viral envelope surfaces, localized exclusively to infected cell plasma and viral envelope lipid bilayers. Distinct from other viral envelope glycoproteins, U22 relies on extracellular glycosylated ectodomains to engage host immune cell surface receptors, while simultaneously modulating chemokine signaling pathways to redirect leukocyte recruitment for sustained viral replication. Under early infection stages, low-level U22 expression mediates subtle immune cell recruitment without robust inflammatory clearance responses. Upon viral late gene activation, abundant U22 envelope glycoprotein drives targeted immune cell chemotaxis to expand susceptible host cell pools and facilitate viral dissemination. Unlike structural viral glycoproteins solely mediating cell entry, U22 bears specialized immunomodulatory immune evasion functions that cannot be fully compensated by other viral glycoproteins, linking viral envelope presentation directly to host leukocyte recruitment and persistent infection pathways. Loss of functional U22 glycoprotein blunts viral capacity to remodel local immune microenvironments and restricts viral tissue spread, while sustained U22 expression suppresses anti-viral innate immune activation, establishing U22 as a core research target for viral glycoprotein immunology and anti-herpesvirus compound screening.

U22 Protein Function: Core Roles in Host Receptor Binding and Viral Immune Evasion Tuning

The biological functions of U22 glycoprotein are focused on glycan-dependent host receptor engagement, leukocyte chemotaxis modulation and anti-viral immune suppression:

  • Glycosylated Host Receptor Binding: Utilizes extracellular glycan domains to interact with immune cell surface chemokine receptors.
  • Immune Cell Chemotaxis Regulation: Redirects myeloid and lymphoid cell recruitment to sites of viral infection.
  • Basal Viral Immune Stealth: Maintains low-level immune remodeling during early infection to avoid rapid host clearance.
  • Anti-Viral Immune Modulation: Governs the magnitude of local inflammatory responses triggered by viral replication.
  • Disease Relevance: U22 loss restricts viral tissue spread, while sustained glycoprotein expression sustains persistent viral infection.
