Background

Structural characteristics and physiological immune function: Protein 4 containing V-set immunoglobulin domain (VSIG4) is encoded by VSI G4 gene and belongs to immunoglobulin superfamily I transmembrane immune receptor, which is specifically expressed in the lipid bilayer structure of cell membrane of macrophages where tissues reside. There are obvious differences between this protein and secretory soluble immunomodulator. Its extracellular domain contains conservative V-set domain and immunoglobulin-like binding domain, and its intracellular terminal sequence is short, and it does not have the catalytic function structure of endogenous kinase, so it is impossible to independently start intracellular continuous signal transduction. As a membrane-anchored immunoregulatory molecule on the surface of macrophages, VSIG4 can specifically bind to complement-derived molecular fragments in physiological steady-state environment, and participate in regulating the immune response of adaptive lymphocytes. When the expression of the membrane localization regulatory factors on the surface of macrophages is insufficient, the body is prone to immune disorders such as overactivation of complement cascade reaction and abnormal activation of lymphocytes, and VSIG4 can play a good role in immune buffering regulation and maintain the cross balance between innate immunity and adaptive immunity in different tissue microenvironment. Due to the heterogeneity of complement components and immune cell composition in different tissue regions, the multi-cell immune system needs to rely on a variety of membrane receptor systems on the surface of macrophages to maintain local immune homeostasis, in which membrane-located VSIG4 can continuously bind complement-related substrates and functional complexes on the surface of immune cells, effectively inhibit abnormal inflammatory reactions and maintain the stability of local immune environment in tissues.

Mutation effect, regulatory mechanism and research value: Mutation of VSIG4 gene can change the binding affinity between receptor and substrate, and then induce the reconstruction of local immune response pattern. Among the macrophage-restricted transmembrane receptors, only VSIG4 has dual core functions of complement fragment recognition and adaptive lymphocyte regulation, and no other similar membrane receptors can completely replace its biological characteristics. The expression level of VSIG4 is highly consistent with the functional activation state of macrophages, which is the key research target to explore the function of immunoglobulin superfamily transmembrane receptor and explain the mechanism of macrophage-mediated immune regulation. At the level of molecular regulation, VSIG4 is stably embedded in the lipid bilayer of cell membrane, and immune regulation is realized by binding to the downstream target molecule complex, and this process will not induce continuous intracellular signal cascade activation. Compared with soluble immune media, the transmembrane localization of its cell membrane can regulate the complement-dependent innate immune response by cell contact, and at the same time mediate the reversible adaptive immune fine-tuning process. The functional defect or down-regulation of VSIG4 will directly interfere with the body's recognition process of complement substrate, significantly weaken the local immune response buffering capacity, and fully highlight the important scientific research value of this gene in the basic research field of macrophage-related transmembrane receptors.

VSIG4 may interact with complement fragments to facilitate pathogen phagocytosis, dampen complement‑ and macrophage‑driven inflammatory responses, and mediate cell‑contact‑dependent suppression of activated T‑lymphocytes.1

VSIG4 Protein Function: Core Roles in Complement-Fragment Recognition and Tissue Immune Coordination

The biological functions of transmembrane VSIG4 immune-regulatory receptor protein are focused on sustained extracellular substrate engagement and tissue immune-response coordination:

  • Broad Molecular-Substrate Affinity: Might interact with multiple complement-derived molecular fragments without triggering consistent intracellular signal cascades. The membrane-bound receptor binds substrate components originating from extracellular tissue compartments and expands the scope of macrophage-dependent immune regulation within tissue microenvironments.
  • Tissue Immune-Homeostasis Regulation: Could moderate excessive complement-cascade turnover and lymphocyte-activation outputs to ease local immune-response overload. This regulatory mode prevents drastic immune-activity fluctuation that disrupt stable tissue physiological conditions.
  • Macrophage-Surface Mediator: Appears to facilitate reversible molecular attachment between VSIG4 extracellular domains and complement-related molecular assemblies. Weak non-covalent receptor-substrate binding generates transient interaction patterns detectable via standard laboratory analytical workflows.
  • Local Immune-Signal Gradient Modulation: Shapes local receptor-substrate interaction gradients to coordinate overall innate-adaptive immune-response intensities.
  • Research Model Relevance: Sequence variants of VSIG4 may shift receptor-substrate binding efficiency within laboratory research systems.

VSIG4 Protein Product

Creative Biolabs offers purified VSIG4 membrane samples produced under unified preparation workflows, including full-length VSIG4 constructs and isolated extracellular immunoglobulin-domain variants. Truncated extracellular fragments cannot support complete receptor-substrate-engagement behaviours, while full-length constructs suit research focused on complement-fragment recognition and macrophage-surface receptor functional observation. All batches receive uniform quality screening. Functional relevant observation may only be carried out with full-length samples under simulated membrane microenvironment setups. Consistent structural features are preserved across batches to support comparative laboratory analysis across separate test groups. Full-length VSIG4 samples retain intact extracellular substrate-binding motif conformation after standardized purification, which supports reliable detection of weak and transient receptor-substrate contacts for comparative functional analysis.
