K-NK: Inhibitory KIR (iKIR) Knockout NK Cell Engineering Service

Natural killer (NK) cell education requires ligands to bind to inhibitory NK cell receptor (iNKR), which is very important for producing self-tolerant functional NK cells. The main family of inhibitory NK cell receptors is inhibitory KIRs (iKIRs), which interact with their HLA class I ligands to play an important role in NK cell regulation and protect normal tissues from NK cell attack. To enhance the anti-tumor efficacy of NK cells, we optimized these unique activated NK cells, which lack KIR inhibitory receptors while retaining a wide range of activated receptors that enable selectively target and kill tumor cells.

Our Unique K-NK Platform

The panoply of activating and inhibiting receptors on NK cells and the myriad mechanisms by which NK cells mediate cytotoxicity provide ample opportunities to engineer NK cells using genetic methods aimed at making signal balance tend to be activated. Our unique NK cell platform has been produced and used as a cell-based "off the shelf" therapy in clinical trials and has proven its potential to kill cancer.

Unlike normal NK cells, our unique K-NK cells do not express iKIRs, which are often used by diseased cells to escape the killing function of NK cells. These unique activated NK cells are optimized to retain a wide range of activated receptors while lacking KIR inhibitory receptors, which can selectively target and kill diseased cells. K-NK cells also carry larger granzymes and perforin-containing particles, allowing them to provide a larger payload of lethal enzymes to multiple targets. Encouraging evidence of safety, mobility and prolonged survival have been proven.
