ELISA is a widely used solid-phase screening method that includes direct, indirect, competitive, and sandwich assays. Double antibody sandwich ELISA is the most commonly used method, while direct and competition ELISA are less commonly used. Sandwich ELISA systems can use either monoclonal or polyclonal antibodies as capture and detection antibodies. Monoclonal antibodies are highly specific for individual epitopes, allowing for precise detection and quantification of small differences in antigens. Polyclonal antibodies are typically used as capture antibodies, while monoclonal antibodies are used as detection antibodies to provide higher specificity. Recombinant monoclonal antibodies can also be used in ELISA, which are less prone to cell line drift or batch-to-batch variation, allowing for optimal antigen specificity.
The ELISA method is widely used for high-throughput antibody discovery and is a variant of affinity assays that aims to establish a positive correlation between ELISA signal values and sample affinity magnitude. However, ELISA may not detect antibodies that recognize target conformational epitopes. Therefore, researchers are encouraged to perform secondary screening techniques such as protein blotting, spot blotting, histology, immunoprecipitation, or radioimmunoassay on candidate ELISA positives.
LFIA is also known as a lateral flow assay (LFA) or a lateral flow test (LFT). It is a classical rapid detection technique based on antigen and antibody immunoassays. LFIA is widely used in biomedical, agricultural, food, and environmental science fields due to its low cost, simplicity, rapidity, and portability.
While the physical composition of the test strips, construction techniques, and buffers play an important role in optimizing the test, at the heart of these processes are the antibodies. Antibodies for LFIA need to be carefully designed and highly purified to ensure a stable supply of antibodies with proven affinity and specificity. Monoclonal antibodies (mostly from mouse hybridomas) are best for large-scale production of specific antibodies. In the process of obtaining antibodies using hybridomas, antibodies need to be screened and validated to ensure that they can meet the needs of LFIA experiments. When screening LFIA-positive hybridomas, clones should be screened by ELISA and WB first. ELISA&WB-positive clones are more likely to be LFIA-positive clones.
