The chemical enzymatic method uses endoglycosidase-catalyzed transglycosylation to link the intact oligosaccharide to the polypeptide in one step, without the participation of any protective group in this process. Enzymatic reaction intermediates as donor substrates can expand the utilization of substrates and also significantly improve the efficiency of enzymatic transglycosylation. In addition, the discovery of some new mutants with sugar synthase-like activity facilitated the synthesis of glycoproteins with full-size natural N-glycans. These advances further prove the important role of chemical enzymatic methods in glycoprotein synthesis.
