In Vitro Protein Evolution by Ribosome Display

Protein Evolution by Ribosome Display

Ribosome display is a cell-free system for in vitro selection and evolution of protein and antibody from large libraries. It uses the principle of coupling individual nascent proteins (phenotypes) to their corresponding mRNA (genotypes), through the formation of stable protein-ribosome-mRNA (PRM) complexes. Compared to other cell-based systems, such as phage display, ribosome display has two important advantages for identifying and optimizing proteins. Firstly, the diversity of ribosome display libraries (1013~1014) is not limited by the transformation efficiency of bacterial cells, which allows the construction of a larger library. Secondly, random mutations can be introduced easily into the selected DNA pool in subsequent cycles, which allows facile directed evolution of binding proteins over several generations. Now a number of studies have been confirmed the potential of ribosome display for directed in vitro protein evolution. Hanes et al. (2000) improved the affinities of a series of scFv antibodies by 40 times by ribosome display technology. Jermutus et al. (2001) demonstrated that the affinity and stability of scFvs can be rapidly and efficiently evolved by ribosome display.
