In Vitro Selection for Enzymatic Activity

Recent advances in library design, and methods of random mutagenesis, combined with new screening and selection tools, provide a powerful strategy to evolve improved biocatalysts. In ribosome display technique, mRNA is not covalently attached to the protein. Thus, the mRNA can be simply eluted. Moreover, the diversity of the ribosome display library is approximately 1012~14, higher than cell-based display libraries (i.e. phage display). The random mutations can be introduced easily after each selection round, allowing the directed evolution of binding proteins over several generations. Amstutz et al. (2001) have reported the in vitro selection for catalytic activity based on catalytic turnover using ribosome display. They designed and synthesized a mechanism-based inhibitor of β-lactamase which is appropriate for the selection of catalytic activity of the ribosome-displayed β-lactamase. Under appropriate selection conditions, an active RTEM-β-lactamase was successfully enriched over an inactive mutant.
