Introduction of SV2B

Synaptic vesicle glycoprotein 2B is a protein that in humans is encoded by the SV2B gene, which is mapped to the chromosome 15q26.1. SV2s are hydrophobic proteins with 12 potential transmembrane regions and a cytoplasmic N- and C-terminus, amongst, the N terminus of SV2s contains a long sequence that varies among the three SV2 proteins but includes a short-conserved region which reacts with the monoclonal SV2 antibody in all three SV2s.

Function of SV2B Membrane Protein

SV2 proteins may modulate synaptic vesicle exocytosis through direct interaction with synaptotagmin 1. SV2B is mainly released in neurotransmitters due to its involvement in the regulation of synaptic vesicles and extracellular secretion, which is widely distributed in the brain. In contrast to SV2A and SV2C, SV2B lacks a significant portion of the cytoplasm, so it can regulate protein interactions. SV2B also lacks regions of the large lumenal domain between transmembrane domains 7 and 8 that are present in SV2A and SV2C. The decrease of SV2B mRNA was already detected before the onset of proteinuria in puromycin aminonucleoside nephropathy (PAN). The mRNA expression of SV2B clearly is altered not only in PAN but also in another proteinuric state that is caused by an antibody against nephrin.

    This article shows that SV2B has an essential role in the formation and maintenance of the glomerular capillary wall.

    This article reveals that SV2B could be a key modulator of amyloid toxicity at the synaptic site.

    This article suggests that the rearrangement produces a fusion of the RUNX1 and SV2B gene antisense strands, a novel translocation partner of RUNX1, which produces an out-of-frame mRNA chimeric transcript and putative truncated RUNX1 forms.

    The authors find that the increase of SV2A expression in the hippocampus at P7 is mainly observed in the CA1 region while SV2B expression in this region remains stable.
