C-C motif chemokine ligand 1 (CCL1) is a small secreted inflammatory chemokine encoded by CCL1 gene, mainly secreted by activated dendritic cells, cutaneous keratinocytes and tissue-resident macrophages upon allergen stimulation. Distinct from broad-spectrum pro-inflammatory chemokines, CCL1 selectively binds CCR8 receptor expressed exclusively on Th2 lymphocytes and cutaneous regulatory T cells to guide targeted immune cell chemotaxis to inflamed skin and mucosal tissues. Under healthy steady-state immunity, minimal basal CCL1 secretion maintains mild cutaneous immune surveillance without excessive inflammatory infiltration. After allergen exposure or skin barrier damage, CCL1 transcription rises sharply, triggering massive Th2 cell migration to lesion sites and releasing IL-4/IL-13 cytokines that amplify atopic dermatitis, hay fever and asthma symptoms. Its CCR8-specific chemoattractant property cannot be fully substituted by other CC chemokines; neutralizing CCL1 suppresses Th2-driven allergic lesions, while sustained CCL1 overexpression worsens chronic inflammatory skin disorders, making CCL1 an irreplaceable core research target for type 2 immunity and anti-allergy compound screening.
CCL executes secreted soluble chemokine function within tissue interstitial fluid, relying on conserved N-terminal CC chemokine fold to form stable dimers and specifically recognize CCR8 transmembrane receptors on Th2 immune cells. Its narrow cell tropism clearly separates CCL1 from pan-leukocyte chemokines, forming a dedicated signaling axis for type 2 allergic inflammation. CCL1-mediated immune cell trafficking links epithelial allergen sensing to Th2 inflammatory amplification, balancing physiological tissue defense and pathological allergic injury according to barrier integrity status. CCL1 participates in cutaneous atopic inflammation and mucosal allergic airway responses. Loss of functional CCL1 greatly reduces Th2 cell accumulation at inflammatory lesion sites. Therefore, CCL1 represents a pivotal research target for CC chemokine study and atopic disease therapeutic exploration.
