Background

CD207 molecule (Langerin) is a transmembrane C-type lectin receptor encoded by the CD207 gene, uniquely expressed in epidermal Langerhans dendritic cell subsets, localized to plasma membrane and intracellular Birbeck granule membranes. Distinct from other dendritic cell lectins, CD207 contains calcium-dependent carbohydrate recognition domains that bind glycan moieties on pathogens and particulate antigens, mediating endocytic antigen uptake and specialized Birbeck granule formation for antigen processing pathways. Under steady cutaneous conditions, basal CD207 expression sustains low-level environmental antigen sampling without robust immune activation. Upon cutaneous pathogen or allergen exposure, glycan ligand crosslinking drives CD207 internalization and antigen sorting to processing compartments, facilitating adaptive immune priming in skin-draining lymphoid tissue. Unlike broadly expressed C-type lectins on myeloid cells, CD207 carries Langerhans cell-restricted antigen capture functions that cannot be fully compensated by other lectin family members, linking glycan antigen recognition to skin dendritic cell immune priming pathways. Loss of functional CD207 impairs cutaneous antigen sampling and weakens skin adaptive immune responses, while elevated CD207 expression enhances particulate antigen internalization, establishing CD207 as a core research target for C-type lectin biology and skin immune modulator screening.

CD207 executes calcium-dependent glycan-sensing lectin function embedded within Langerhans cell membrane bilayers, utilizing conserved carbohydrate recognition domains to mediate selective calcium-dependent binding of microbial and environmental glycans. Conserved glycan binding pockets restrict substrate recognition to pathogen surface carbohydrate structures, separating its ligand spectrum from other myeloid C-type lectins. CD207-mediated antigen uptake bridges extracellular cutaneous antigen capture and intracellular antigen processing machinery, balancing dendritic cell antigen sampling intensity relative to environmental stimulus load. CD207 participates in Birbeck granule biogenesis, skin pathogen recognition and naive T cell priming. Deficient CD207 expression disrupts Langerhans cell antigen trafficking and impairs cutaneous immune surveillance. Therefore, CD207 represents a pivotal research target for tissue-restricted C-type lectin study and dermatological immune compound screening.

CD207 Protein Function: Core Roles in Calcium-Dependent Glycan Capture and Skin Dendritic Cell Tuning

The biological functions of CD207 are focused on calcium-mediated glycan ligand binding, endocytic antigen uptake and cutaneous immune control:

  • C-Type Lectin Glycan Recognition: Binds pathogen and environmental carbohydrate ligands via calcium-dependent binding pockets.
  • Birbeck Granule Biogenesis: Drives formation of specialized intracellular antigen-processing storage compartments.
  • Cutaneous Immune Steady-State: Maintains low-level environmental antigen sampling under intact skin barrier conditions.
  • Adaptive Immune Priming Modulation: Adjusts efficiency of antigen presentation to naive lymphocytes upon skin pathogen exposure.
  • Disease Relevance: CD207 loss impairs skin antigen surveillance, while intact lectin activity enhances particulate antigen capture capacity.
