C-type lectin domain containing 6A (CLEC6A), also known as Dectin-2, is a single-pass transmembrane C-type lectin receptor encoded by the CLEC6A gene. This protein localizes to the plasma membrane of several immune-related cell populations. CLEC6A contains a large extracellular C-type lectin-like domain, a single transmembrane segment, and a short intracellular cytoplasmic tail that lacks an intrinsic signaling motif. Its extracellular lectin-like domain projects into the extracellular space and contains conserved structural features associated with calcium-dependent carbohydrate recognition. For signal transduction, CLEC6A associates with the Fc receptor γ-chain (FcRγ/FCER1G), whose intracellular ITAM becomes phosphorylated following receptor activation and recruits Syk to initiate downstream signaling. Polypeptide biosynthesis and folding take place within endoplasmic reticulum compartments before surface trafficking, and correct folding of the extracellular lectin-like domain depends on chaperone-assisted quality control processes. CLEC6A exhibits a cell-type-restricted expression pattern, detectable primarily among specialized immune cell subsets, and its transcript abundance can shift according to the differentiation status of immune lineage cells. Polypeptide folding surveillance within secretory compartments helps ensure that properly assembled receptor complexes reach the cell surface to exert receptor-related activities.
The extracellular C type lectin like domain of CLEC6A mediates molecular pattern recognition toward distinct extracellular molecular entities. Calcium ion coordination contributes to maintaining the folded conformation required for target molecule engagement. Upon extracellular ligand engagement, sequence motifs within its short intracellular cytoplasmic segment recruit cytoplasmic signalling adaptor molecules to initiate downstream intracellular signal events. Other C type lectin family receptors share general lectin domain architecture but display divergent molecular recognition preferences defined by differences within their extracellular binding surfaces. Related paralog receptors cannot fully reproduce the molecular recognition profile exhibited by CLEC6A. Changes to CLEC6A surface abundance alter quantity of available pattern recognition receptors on immune cell plasma membrane. Sequence mutation within extracellular lectin like domain may interfere with target molecule binding, while alteration of intracellular motif sequences may disturb recruitment of downstream cytoplasmic signalling adaptor components. Varied receptor abundance can adjust cellular sensitivity toward extracellular molecular patterns without completely abolishing receptor associated response capacity.
Fig. 1 Schematic representation of CLEC6A (Dectin‑2) signalling architecture. The receptor carries an extracellular recognition domain and transmembrane segment. It does not possess intrinsic cytoplasmic signalling motifs and recruits Syk signalling components via accessory adaptor molecules.1
The biological functions of CLEC6A are focused on extracellular molecular pattern recognition, calcium dependent target engagement and intracellular signalling adaptor recruitment:
Creative Biolabs offers purified CLEC6A membrane samples via standardized preparation workflows, including full length CLEC6A constructs and isolated extracellular lectin domain variants. Isolated extracellular lectin domain fragments may not support complete target recognition and cytoplasmic adaptor recruitment related behaviours, while full length constructs may be suited for C type lectin receptor pattern recognition related research. All samples receive routine quality screening, and functional relevant observation may only be carried out with full length samples under simulated membrane environments. All sample batches follow unified processing standards to maintain consistent structural features for comparative laboratory analysis across separate test groups.
