Background

Type C lectin domain protein 9A(CLEC9A) is a type II transmembrane pattern recognition receptor encoded by CLEC9A gene, which is specifically expressed in a subset of classical type 1 dendritic cells (cDC1) and located on the surface of dendritic cell membrane and endosomal vesicles. Unlike other type C lectin receptors that recognize pathogens, CLEC9A can specifically recognize filamentous actin released by necrotic injury cells and tumor cells, and perceive it as endogenous injury-related molecular patterns (DAMPs). After the receptor binds to the ligand, the endocytosis process is started, and CLEC9A transports the internalized tumor components to the cross-presenting endosome, completing the loading of MHC class I molecules of tumor antigens, thus activating the anti-tumor adaptive immunity mediated by CD8⁺ cytotoxic T cells. Under normal tissue steady state, the basic expression level of CLEC9A is low, which can avoid abnormal uptake of its own tissue fragments and maintain peripheral immune tolerance; The actin ligand released by a large number of necrotic cells in tumor microenvironment can continuously activate CLEC9A signal and induce a powerful anti-tumor immune response. Other myeloid lectins can't compensate the cross-presentation function of CLEC9A, which relies on cDC1. Knocking out CLEC9A will significantly weaken the cross-sensitization ability of tumor and accelerate the malignant progress of tumor. The antigen delivery system targeting CLEC9A can greatly improve the effect of tumor immunotherapy, making it the core target of dendritic cell immune mechanism research and tumor vaccine development.

CLEC9A is anchored in the cell membrane of cDC1, which mainly plays the role of injury perception and antigen uptake, and relies on extracellular C-lectin domain to specifically bind cytoskeleton actin exposed by necrotic cells. Its recognition characteristics of endogenous damage ligands are different from those of CLR family members who traditionally perceive microorganisms, and constitute a unique tumor immune monitoring receptor pathway in the body. CLEC9A-mediated endocytosis can link the signal of tissue necrosis with the cross-presentation mechanism of MHC class I antigen, and dynamically regulate the balance between anti-tumor immunity and self-tolerance according to the degree of tissue injury. This molecule is involved in key immune processes such as cross-sensitization of solid tumor antigen and anti-virus CD8 T cell response, and the loss of CLEC9A function will completely block the efficient tumor-specific CD8 T cell activation. Therefore, CLEC9A is the key research target of dendritic cell receptor research and tumor immunotherapy screening.
