Cytokine receptor-like factor 2 (CRLF2), encoded by the CRLF2 gene and also known as TSLPR or CRL2, is a member of the type I cytokine receptor family. The CRLF2 gene is located in the pseudoautosomal region of the sex chromosomes at Xp22.3 and Yp11.3, a region where genes encoding the IL-3 receptor alpha and GM-CSF receptor alpha chains are also situated. The deduced amino acid sequence of 371 residues includes a typical signal sequence and a single membrane-spanning region. The mature protein is predicted to have a molecular mass of approximately 39.7 kDa. Structurally, the N-terminal extracellular region contains two fibronectin type III-like domains, four conserved cysteine residues, and a WSXWS box-like motif—all characteristic features of type I cytokine receptor family members. The C-terminal intracellular region contains box 1 and box 2-like motifs that mediate interactions with JAK kinases. CRLF2 is expressed in heart, skeletal muscle, kidney, and adult and fetal liver, and is preferentially expressed in myeloid cells including dendritic cells and monocytes, with weaker expression in T cells. The WSXWS motif appears to be necessary for proper protein folding and efficient intracellular transport.
CRLF2 functions as the receptor subunit for thymic stromal lymphopoietin (TSLP). CRLF2 heterodimerizes with the interleukin-7 receptor alpha chain (IL-7Rα/CD127) to form the heterodimeric thymic stromal lymphopoietin receptor (TSLPR). Binding of TSLP to CRLF2 is a mechanistic prerequisite for recruitment of IL-7Rα to form the high-affinity ternary complex. Formation of this functional receptor complex activates receptor-associated JAK2 and downstream signaling pathways, prominently including STAT3 and STAT5, which contribute to the regulation of cell proliferation and development of the hematopoietic system. Through its participation in the TSLP receptor system, CRLF2 is involved in processes such as hematopoietic cell development, dendritic cell maturation, and immune cell signaling. Its receptor subunit function makes CRLF2 a valuable research target for studies of type I cytokine receptor biology, JAK-STAT signaling, hematopoietic development, and immune regulation.

Creative Biolabs offers high-quality CRLF2 membrane protein products produced using optimized expression systems. These include full-length type I transmembrane protein that retains the native membrane-spanning domain and cytoplasmic box 1/box 2 motifs, as well as isolated extracellular domain variants containing the fibronectin type III-like domains and WSXWS box-like motif. The extracellular domains mediate TSLP binding and heterodimer assembly with IL-7Rα, making these proteins suitable for studies of TSLP receptor-ligand interactions, heterodimer formation, and JAK-STAT signaling characterization. All CRLF2 proteins undergo strict quality control to support consistent performance across applicable research platforms.
