Presenilin enhancer, gamma-secretase subunit (PSENEN), also known as PEN-2, is a small integral membrane protein of approximately 101 amino acids and an essential component of the γ-secretase complex. Together with presenilin, nicastrin, and APH1, PSENEN contributes to the assembly and maturation of the functional γ-secretase complex. Presenilin contains the catalytic aspartate residues responsible for intramembrane proteolysis, whereas PSENEN plays an important role in stabilizing the complex and promoting presenilin endoproteolysis into its N-terminal and C-terminal fragments. Loss or disruption of PSENEN can impair γ-secretase complex maturation and reduce γ-secretase-mediated substrate processing.
