Transforming growth factor-alpha (TGFA), or EGF-like TGF, is a potent mitogenic polypeptide of the epidermal growth factor (EGF) family of cytokines. TGFA transitions through a transmembrane precursor (pro-TGF-α) that is cleaved to release a soluble bioactive form, while the membrane-associated precursor can also support juxtacrine signaling, allowing TGFA to act through autocrine, paracrine, and juxtacrine mechanisms. The mature protein has ~40% sequence homology with EGF, competes directly for EGFR binding, but displays unique biological properties that are important for tissue development and repair in physiological settings as well as pathological processes such as oncogenesis. In mammals, TGFA has a wide tissue distribution including the pituitary gland, keratinocytes, macrophages, and multiple other epithelial cell populations, while expression patterns are tightly regulated during embryogenesis and tissue repair. Dysregulated TGF-α/EGFR autocrine or juxtacrine signaling can support tumorigenicity and has been implicated in multiple forms of malignant neoplasm by maintaining chronic proliferative signaling. Therefore, TGFA has become an interesting target and a useful tool for basic research in cancer biology, regenerative medicine, and developmental biology.

The functional landscape of TGFA is broader than simply mitogenic activation and this extends to diverse physiological and pathological processes:

Generating strong cellular models that express more physiologically relevant levels of TGFA is critical to further mechanistic understanding and therapeutics. Creative Biolabs offers precisely designed TGFA stable cell lines that enable uniform and sustained expression of target TGFA protein from selectable markers. These stable cell lines act as invaluable tools for high-throughput drug screening, ligand-receptor interaction studies, signal transduction pathway mapping and functional genomics research. The validation steps in our cell line development pipeline contain flow cytometric confirmation of surface expression, verification of total protein levels by Western blot and functional assay validation to ensure biological fidelity. From TGFA-overexpression models for oncogenic signaling or reporter cell lines for pathway activation maintenance, our custom-made solutions will speed your research timeline while ensuring high quality experimental reproducibility.

Our comprehensive portfolio of high-affinity recombinant antibodies against TGFA are based on advanced hybridoma and phage display technologies that guarantee high specificity, sensitivity and lot-to-lot consistency. Our recombinant TGFA antibodies differ from traditional polyclonal preparations in that their clonality is defined and they are validated to have low cross-reactivity with other members of the EGF family, while being functional across a wide range of applications including: Western Blotting (WB), ELISA, Flow Cytometry (FCM), Immunofluorescence (IF), Immunocytochemistry (ICC), Immunohistochemistry (IHC) and Immunoprecipitation (IP). Here, we describe a set of highly specific anti-TGFA antibodies that permit the accurate and quantitative detection of both soluble and membrane-bound forms in various biological matrices (e.g., culture medium or tissue extracts), thus enabling investigators to confidently explore TGFA-mediated signaling.

In addition to the catalog capabilities, Creative Biolabs also offers custom protein engineering and antibody discovery services customized to meet the special requirements of TGFA-related research programs:
No—our TGFA products including recombinant proteins, stable cell lines and antibody reagents are developed solely for research use only. All of these have not yet gone through the necessary safety, sterility and regulatory testing for use in humans.
Yes, we generated stable lines with strong surface pro-TGFA expression as verified by flow cytometry using the C-terminal–specific antibody.
Yes, we provide recombinant proteins and stable cell lines expressing the variants with designed mutations within the receptor-binding surface to prevent strong EGFR interactions while maintaining proper folding of the domain region.
Yes, the immunogen is associated with a region that shows high conservation between mammals which allows for detection in mouse, and rat tissue preparations and cell lysates without the need for multiple species-specific reagents.
