Background

Transforming growth factor-alpha (TGFA), or EGF-like TGF, is a potent mitogenic polypeptide of the epidermal growth factor (EGF) family of cytokines. TGFA transitions through a transmembrane precursor (pro-TGF-α) that is cleaved to release a soluble bioactive form, while the membrane-associated precursor can also support juxtacrine signaling, allowing TGFA to act through autocrine, paracrine, and juxtacrine mechanisms. The mature protein has ~40% sequence homology with EGF, competes directly for EGFR binding, but displays unique biological properties that are important for tissue development and repair in physiological settings as well as pathological processes such as oncogenesis. In mammals, TGFA has a wide tissue distribution including the pituitary gland, keratinocytes, macrophages, and multiple other epithelial cell populations, while expression patterns are tightly regulated during embryogenesis and tissue repair. Dysregulated TGF-α/EGFR autocrine or juxtacrine signaling can support tumorigenicity and has been implicated in multiple forms of malignant neoplasm by maintaining chronic proliferative signaling. Therefore, TGFA has become an interesting target and a useful tool for basic research in cancer biology, regenerative medicine, and developmental biology.

TGFA Protein Function: A Central Regulator of Cellular Homeostasis

The functional landscape of TGFA is broader than simply mitogenic activation and this extends to diverse physiological and pathological processes:

  • Epithelial Cell Proliferation and Tissue Regeneration: TGFA is a strong keratinocyte mitogen that stimulates EGFR activity in skin wound healing, corneal epithelial repair and mucosal tissue regeneration.
