Background

Translocase of outer mitochondrial membrane 20 (TOMM20) is a single-pass outer mitochondrial membrane receptor subunit encoded by the TOMM20 gene, serving as a primary precursor recognition component of the multi-subunit TOM translocase complex. TOMM20 contains an N-terminal transmembrane anchor with a short N-terminal segment facing the mitochondrial intermembrane space and a cytosolic C-terminal receptor domain that specifically binds positively charged mitochondrial targeting presequences of nuclear-encoded precursor proteins before guiding them into the central translocation pore formed by TOMM40. Without functional TOMM20, most mitochondrial precursor polypeptides fail to cross the outer membrane and accumulate in the cytosol to trigger cellular stress responses.
