A recent study has showcased the successful application of yeast display for isolating anti-idiotype vNARs targeting specific BCR idiotypes from lymphoma cell lines. The idiotype, a tumor-specific and functionally active BCR, varies from patient to patient, making targeting challenges. Shark-derived antibody domains (vNARs) from a semi-synthetic yeast display CDR3-randomized library were used to generate anti-idiotype binders with strong affinity. The study reports an alternative approach for personalized B-cell lymphoma therapy based on the construction of vNAR-Fc ADCs to enable specific killing of malignant B cells, which may widen the therapeutic window for B-cell lymphoma therapy.
