The baculovirus expression vector system (BEVS) is universally recognized as a powerful and versatile system for the production of high-quality proteins. There are several advantages of insect cells over E. coli such as improved solubility, ability to incorporate post-translational modifications, and higher yields for secreted proteins.
The gene encoding the protein of interest is cloned into a suitable transfer vector, such as pFastBac1. This vector contains elements necessary for the expression of the gene in insect cells. The transfer vector is co-transfected with baculovirus DNA into insect host cells to obtain recombinant baculoviruses containing the gene of interest.
The recombinant baculovirus is amplified through multiple rounds of infection and harvesting to ensure that a sufficient quantity of recombinant virus is available to infect insect cells at a large scale.
The expressed protein is purified using affinity chromatography. The choice of purification method depends on the protein's properties and the presence of any affinity tags that were included during gene cloning.
The final product undergoes quality control tests to verify its purity, activity, and structural integrity. We can provide quality control methods including SDS-PAGE, WB, ELISA, and so on.
