Background

C-X-C motif chemokine receptor 1 (CXCR1) is a class A rhodopsin-like G protein-coupled receptor (GPCR), which is predominantly couples with heterotrimeric Gi proteins upon agonist engagement, leading to phospholipase C activation, intracellular calcium mobilization, and downstream activation of mitogen-activated protein kinase (MAPK) pathway and phosphoinositide 3–kinase (PI3K)/AKT pathways. In homeostatic environments, CXCR1-mediated trafficking is required to rapidly contain pathogens and repair damaged tissues. However, in chronic disease, dysregulated CXCR1 signalling promotes tissue destruction in acute respiratory distress syndrome and neutrophil extracellular trap formation in autoimmune vasculitis, driving the recruitment of protumoural neutrophils and myeloid-derived suppressor cells into the breast, pancreatic, and colorectal carcinoma tumour microenvironment. The clinical validation of CXCR1 as a pharmacologically tractable target, bolstered by preclinical efficacy of receptor antagonists in inflammatory and oncological models, has created greater need for structurally authentic ligand-free receptor preparations, pharmacologically faithful cell lines and conformation-specific antibodies to inform modern drug discovery.

CXCR1 Protein Function: A Chemotactic Command Center for Neutrophils and Beyond

CXCR1 has a functional reach across multiple interrelated physiological and pathological landscapes:

  • Commander of acute inflammation: CXCR1 sets sharp chemotactic gradients which direct blood neutrophils into tissues infected or injured within minutes from insult (h) That its ability to elicit both rapid morphogenesis and long-lasting directional migration, guarantees that microbial invaders are met with a powerful phagocytic-oxidative arsenal upon the first line of innate immune defense.
  • Tumor Microenvironment Sculptor: CXCR1 expression on neutrophils and myeloid-derived suppressor cells (MDSCs) within malignant tissues enables targeted recruitment towards tumor-derived CXCL8, followed by secretion of arginase, reactive oxygen species (ROS), matrix metalloproteinases which inhibit the function of cytotoxic T-cells whilst fostering metastatic seeding. Pharmacological blockade of this axis reorients the tumor microenvironment to an immune permissive state.
  • Therapeutic Biomarker & Drug Target: High circulating CXCL8 plasma levels and higher tumor-infiltrating neutrophils expressing CXCR1 are associated with poor prognosis in many solid malignancies and resistance to checkpoint inhibition. New precision intervention tools are advancing through preclinical pipelines in the form of small-molecule antagonists and monoclonal antibodies that block CXCR1-mediated signaling for inflammatory disorders and combination immuno-oncology strategies.
