Murine Hybridoma Introduction

Antibody production from hybridoma has been found widespread use in various fields. Monoclonal antibodies prepared from murine hybridoma offer several advantages, including a stable source, easy preparation, and high yield. As a result, they are commonly used for immunodetection analysis and early disease screening. Preparing murine hybridoma monoclonal antibodies involves several steps, such as antigen design and screening, animal immunization, cell fusion screening technology, antibody purification, among others.

Conventional Murine Cybridoma Fusion Technique

The general procedure for mouse hybridoma preparation involves antigen preparation and identification, animal immunization with the antigen, and determining the serum titer of the immunized animals. Once the titer has reached the required level, the spleen cells and myeloma cells are fused. Hybridoma cells are then screened using HAT and HT selective media and indirect ELISA to determine if the resulting hybridomas can both proliferate indefinitely and secrete hybridoma antibodies specific to the antigen.

Hybridoma Fusion Techniques in Transgenic Murine

In transgenic technology, the antibody gene encoding humanized antibody is transferred into the antibody gene null mice. Depending on the transferred genes, the hybridomas prepared from transgenic mice can be divided into chimeric antibodies and fully humanized antibodies. A chimeric antibody is an antibody molecule composed of the variable region gene of a murine antibody and the constant region gene of a human antibody through DNA recombination, assembled with a vector and transfected into cells for expression. The transgenic technology reduces the mouse-derived components, thus reducing the rejection caused by mouse-derived antibodies and helping to improve efficacy. Fully humanized antibody refers to the transfer of all human antibody coding genes into genetically modified antibody gene-deficient mice through genetic recombination technology, allowing the mouse expression system to fully express human antibodies.
