Rat Hybridoma Introduction

Rat hybridoma as an important application of hybridoma technology is another antibody production from hybridoma developed after the mouse hybridoma technology and offers unique advantages. Rat hybridoma has a wide antigen recognition spectrum and produces high yields of rat monoclonal antibodies. Additionally, it and can be used for the study of mouse antigens, which complements the mouse hybridoma technology.

The necessity of Rat Hybridoma

The use of rat hybridoma models to produce monoclonal antibodies is necessary due to the limitations of using mice to establish hybridoma models. Although the technology of producing monoclonal antibodies using mice is mature, it still has some limitations that rat hybridoma can address. Firstly, the antibody repertoire in rats differs from that in mice. For instance, rats are better suited for studying xenoantigens of mouse species using rat mabs, and they are better for studying rat alloantigens. Additionally, rats tend to exhibit stronger immune responses to some non-rat and mouse-derived antigens than mice.

Secondly, the production of rat monoclonal antibodies in vivo is more efficient compared to mice. Although rats are about ten times larger than mice, both species have similar gestation periods of approximately three weeks, and they reach adulthood at around 10-12 weeks of age. Therefore, with the same incubation time, rats produce more antibodies, and the process is easier and less costly.

Finally, rat monoclonal antibody purification methods are established and convenient to use. The rat mab purification method is fast, efficient, and inexpensive. Both the in vitro culture supernatant method and ascites in vivo can be used for the purification of rat monoclonal antibodies.
