Screening in Action: Case Studies and Results

Target Protein Binding Peptide Screening using TriCo-16 and TriCo-20 libraries
Objective To rapidly identify novel peptide sequences with high-affinity binding to a specific target protein using our TriCo-16 and TriCo-20 phage display peptide libraries.
Strategy We conducted a comprehensive four-round screening process using both the TriCo-16 and TriCo-20 phage display peptide libraries. Following screening, preliminary validation and deep sequencing were performed using small-scale phage ELISA to monitor the binding activity and enrichment efficiency.
Key Results 1. Initial Discovery & Enrichment
Through the iterative four-round screening and preliminary phage ELISA validation, we initially identified 3-5 promising target peptide sequences. This initial success demonstrated the effectiveness of our screening protocol in enriching for binders.

2. Deep Sequencing for Comprehensive Discovery
To maximize the discovery of potential peptide binders and to analyze their binding motifs comprehensively, we performed Next-Generation Sequencing (NGS) on the enriched phage pools. The NGS analysis revealed that the top 10 most abundant sequences accounted for approximately half of the total abundance, indicating a strong enrichment of specific binders.

3. Synthesis and Validation of Top Binders
The identified top 10 peptide sequences from the NGS analysis were subsequently synthesized for further activity validation. All top 10 synthesized peptides demonstrated strong positive binding to the target protein, confirming their high affinity and specificity.

This project successfully identified multiple high-affinity peptide binders for the target protein by combining efficient phage display screening with advanced NGS analysis. Our ability to not only isolate initial candidates but also deeply characterize the enriched library to uncover a diverse set of strong binders, followed by successful synthesis and validation, underscores our comprehensive service capabilities in peptide discovery and development.
