Case: Screening for Anti-Type 1 Idiotype Antibody

After conducting four rounds of screening and validating 92 randomly cloned phages through ELISA, we successfully obtained nine candidate clones that specifically target the antigen. These candidates were further validated for their anti-idiotype properties using soluble competitive ELISA techniques.

We conducted four rounds of screening targeting the MHC-peptide. After identification through monoclonal phage ELISA, we obtained seven positive clones. These clones were further validated for their binding capacity using soluble ELISA. All seven clones exhibited specific bindings to the target MHC-peptide and did not bind to the control MHC-peptide.

We screened antibodies that can specifically bind to cell-internalizing targets using phage display technology. The internalization process was validated through flow cytometry, demonstrating that most screened antibodies were able to efficiently enter the target cells.
