A therapeutic antibody's antigen-binding site, located in its variable region, contains a unique set of amino acid sequences known as the idiotope (Id). This idiotope is itself immunogenic and can elicit an immune response, leading to the generation of anti-idiotype antibodies (Anti-ID Abs). These specialized antibodies are critical reagents because they specifically recognize the unique idiotope of a particular antibody drug, making them indispensable tools for ligand-binding assays (LBAs) that quantify the drug or detect an immune response against it.
