Antibody affinity. (Creative Biolabs Authorized).

The Critical Importance of Accurate Affinity Measurement (Kᴅ)

Antibody affinity, quantified by the equilibrium dissociation constant (Kᴅ), is inversely related to the binding strength. A low Kᴅ value signifies a high-affinity interaction, meaning the antibody binds tightly to its target and will likely remain bound longer, which is often a desirable trait for therapeutic efficacy.

Why is precise Kᴅ determination essential?

Our State-of-the-Art Antibody Affinity Measurement Platforms

At Creative Biolabs, we believe a multi-platform approach is key to generating the most reliable and relevant data. No single technology is optimal for every antibody-antigen pair. We tailor our strategy to your specific molecule, matrix, and research question, utilizing an unparalleled suite of cutting-edge instrumentation.

Surface Plasmon Resonance (SPR)

SPR is a label-free, real-time optical technique that measures the binding of an analyte flowing over a ligand immobilized on a sensor chip. It is the industry's gold standard for its precision and the wealth of kinetic data it provides.

Bio-Layer Interferometry (BLI)

BLI is another powerful, label-free optical technique that measures the interference pattern of white light reflected from two surfaces: a layer of immobilized protein on a biosensor tip and an internal reference layer.

A Comprehensive Suite of Affinity Measurement Services

Our expertise covers a wide array of applications and molecule types.

Service Category Description Key Applications
Full Kinetic & Affinity Analysis Provides comprehensive kinetic profiles (kₐ, kₑ) and high-precision equilibrium affinity (Kᴅ) using SPR or BLI. Lead optimization, MoA studies, IND-enabling studies.
High-Throughput Affinity Screening Rapidly screens hundreds or thousands of antibody candidates from hybridoma supernatants or phage display outputs to identify and rank high-affinity binders. Early-stage discovery, antibody library screening.
Epitope Binning & Mapping Groups a panel of monoclonal antibodies based on their ability to simultaneously bind to an antigen, defining their epitope landscape. Patent claims (IP), selecting antibody pairs for diagnostic assays, understanding MoA.
Complex Interaction Analysis Specialized assays designed to measure the affinity of challenging interactions. Small molecule drug discovery, bispecific antibody characterization, vaccine development.
Affinity in Complex Matrices Leverages the power of BLI and flow cytometry to measure binding directly in crude samples like serum, plasma, or cell lysates. Preclinical studies, pharmacokinetic/pharmacodynamic (PK/PD) modeling.
Comparability & Biosimilar Studies Performs side-by-side affinity and kinetic analysis to demonstrate biosimilarity between a reference product and a biosimilar candidate, a critical regulatory requirement. Biosimilar development, manufacturing process changes.

We can efficiently identify antibody isotypes using Biacore, ProteOn and Octet systems. For instance, when Octect system is applied, the biosensor immobilized with an certain type of anti-species antibody will capture a specific isotype of antibody from a crude sample, such as a hybridoma supernatant. Therefore, identifying the isotype of a specific antibody is achieved.
