Antigen Preparation
The client provided information and materials for a small molecule hapten. Based on the data provided and project objectives, we prepared the antigen material for both immunization and subsequent screening.

We immunized three rabbits and continuously monitored their immune titers. After several rounds of testing, we selected the two rabbits with the most robust immune responses for further use in library construction.

Using peripheral blood mononuclear cells (PBMCs) and spleen cells from the two selected rabbits, we constructed a phage display Fab library. The library met all quality control standards, ensuring a high diversity of antibody candidates for the screening process.

We performed 4 rounds of screening using conjugated small-molecule antigens. Effective enrichment of binders was observed, confirming the presence of high-affinity antibodies specific to the target antigen. The screening process was optimized to identify the most promising candidates.

Phage-based assays were used to validate the binders. We sequenced all positive clones and identified approximately 5 unique antibody candidates. These sequences were delivered to the client, providing them with a selection of high-potential antibodies for further development.
