Case Highlights: Transforming Research with Antibody Libraries and Screening

Sheep Immune Antibody Library Construction and Screening of Target Protein
Objective
The client provided immunized sheep samples. Our goal was to construct an immune antibody library and perform library screening to identify specific binders. We also carried out antibody expression and further characterization to validate the quality and functionality of the selected antibodies.
Summary
We successfully constructed high-quality Fab libraries, identified and validated high-affinity binders, and produced functional chimeric antibodies, providing the client with valuable tools for further research and therapeutic development.
1. Sample Preparation and Library Construction
Immunized sheep samples were provided. Sheep antibody genes were amplified and cloned into best-fit phagemid vectors.
Library QC Results:
1×109 Large Capacity 100% Insertion rate Highly Diversity
2. Library Screening
Three rounds of phage display screening were performed, leading to significant enrichment of binders from the sheep library. A clear differentiation between target and control groups was observed.

3. Binder Validation
Phage ELISA Testing: 25 clones binding positively to the target protein.
Sequencing: 18 unique clones from the sheep library were identified, confirming diversity and specificity.
Soluble Expression: 13 clones expressed soluble antibodies, all showing strong binding to target protein.

4. Antibody Pilot-Test Production
Expression in E. coli system: Four clones were successfully expressed, yielding >1 mg.
ELISA Results: All antibodies exhibited strong binding activity, with EC50 values below 1 nM for most clones.
5. Sheep Chimeric Antibody Production
Chimeric versions of selected clones showed comparable binding activity to full sheep antibody.
