Allergic Diseases Vaccines

Allergic diseases are prevalent worldwide. Allergen Immunotherapy (AIT) is a current treatment for allergic diseases, leading to modification of the natural course of the disease. Vaccine-based immunotherapy is considered to be a more effective, convenient, and safer option with promising advances in recent years. Creative Biolabs is a world leader in the field of vaccine development. With our extensive experience and advanced platform, we are therefore confident in offering the best vaccine development services for allergic disease.

Background of Allergic Diseases

Vaccines for Allergic Diseases– Creative Biolabs

Allergic diseases are one of the most prevalent diseases in the world, affect approximately one-third of the world’s population and the prevalence continues to rise. Allergic diseases are characterized by an inappropriate immune response to common environmental allergens, the most common of which are derived from food, animal fur, plant pollen, and latex. Currently, patients are limited to avoiding and using readily available emergency medications, but these methods have certain limitations. In recent years, immunotherapy has been recognized as an attractive therapeutic strategy to induce tolerance. To date, a variety of allergens have been identified and used for immunotherapy of allergic diseases. Studies of natural allergens have demonstrated the efficacy of immunotherapy for allergic diseases. Advances in molecular biology techniques, including purification, sequencing, and cloning, enable researchers to identify specific allergen components and T cell binding epitopes. By manipulating allergen genes or structures and IgE binding, allergenicity can be reduced, thereby reducing systemic responses, and making recombinant DNA and peptide vaccines a safe and effective form of immunotherapy.

DNA Vaccines

Vaccination with allergen-encoding bacterial plasmid DNA (pDNA) is an interesting strategy that is designed to stimulate an immune response against a particular protein. In DNA vaccines, DNA encoding allergen is administered in bacterial plasmid vectors, which express the allergen epitope. Allergen DNA vaccination involves the intramuscular or intradermal injection of pDNA encoding allergens, resulting in in vivo transfection of muscle cells and/or antigen presenting cells (APCs) that subsequently express and initiate an allergen-specific Th1 immune response. Since allergens produced by the host are intracellular and secreted only in small amounts, this type of allergen vaccination does not cause significant allergic and anaphylactic reactions. Furthermore, this Th1 immune response to allergens is superior to pre-existing allergic Th2 responses. Therefore, as a novel, safe, and effective allergen-specific immunotherapy, DNA vaccine has great potential in the prevention and treatment of allergic diseases.

Peptide-Based Vaccine

Peptide immunotherapy is allergen immunotherapy based on a modified allergen or peptide fragment. Peptide-based foods or other allergic vaccines contain short, synthetic, allergen-derived peptides that correspond to T cell epitopes from different allergens. T cell epitopes are peptide sequences that are introduced to antigen presenting cells, which are capable of inducing immunologic tolerance. The modified peptide vaccine involves an allergen protein that has undergone point mutations of key amino acids, in order to alter the epitopes that are responsible for binding IgE. Modification of these IgE binding epitopes decreases the affinity of this interaction and should, in theory, enhance the safety of this approach.
