Neuroblastoma in children

Although acute lymphoblastic leukemia is most common and curable among cancers in children, metastatic solid tumors are often fatal. The expression of major histocompatibility complex (MHC) is often low or absent in pediatric solid tumors, including neuroblastoma, Ewing sarcoma, and alveolar rhabdomyosarcoma, and is accompanied by overexpression of immune inhibitory molecules like programmed death ligand 1 (PD-L1) and B7-H3. In addition to the immunosuppressive tumor microenvironment, the neoantigen landscape based on mutation load is far more restricted among pediatric tumors (less than 10 mutations per tumor), thereby limiting the potential actionable repertoire of immune targets. Furthermore, these cancers typically affect young children when the innate and adaptive immune systems are naive, with little time to enrich tumor specific adaptive immunity. Partly because of the limited repertoire of neoantigens, available targets for immunotherapy in children are seldom tumor specific and are often directed at differentiation antigens leading to autoimmune side effects—a consideration of particular relevance if cured patients are expected to live for decades.

Neuroblastoma

Neuroblastoma is a cancer which forms in certain types of nerve tissue. About 90% of cases occur in children under 5 years old and it is rare in adults. Of cancer deaths in children, about 15% are caused by neuroblastoma. It most frequently starts from one of the adrenal glands, but can also develop in the neck, chest, abdomen, or spine.

Leukemia

Leukemia is a type of cancer arising from the bone marrow and causes a large number of abnormal white blood cells. These white blood cells are not fully developed and are known as blast cells or leukemic cells. Symptoms may include fever, bruising and bleeding, feeling tired and increased risk of infection. These symptoms are caused by the lack of normal blood cells. Diagnosis is usually done by blood test or bone marrow biopsy.

Lymphomas

Lymphoid malignancies represent the largest class of hematologic cancers and are highly heterogeneous in their histologic characteristics and clinical behavior. It is a group of hematological cancers that develop from lymphocytes, a type of white blood cell. There are dozens of subtypes of lymphomas. The two main categories of lymphomas are Hodgkin's lymphomas (HL) and the non-Hodgkin lymphomas (NHL).

Brain Tumors

Malignant brain tumors are classified into primary and secondary (metastatic) brain tumors. Primary brain tumors can arise from any tissue in the central nervous system and are classified into benign and malignant tumors. Secondary (metastatic) brain tumors are a common complication among patients with systemic cancer and more common than primary brain tumors.
