Characteristics of Controlled Release Vaccines

The attributes of this "perfect" vaccine technology include:

  • Consisting of a vaccine that provides complete and long-term protection after a single immunization;

  • Universal use of this technology for most existing vaccines.

Single Administration Vaccine (SAV)

Researchers have studied the possibility of developing single-dose vaccine (SAV). The main methods include:

  • Viral vectors encoding all or part of the pathogen genome;
  • DNA vectors carrying genes of pathogen: DNA vectors for immunization have recently attracted a great deal of attention as they are capable of producing humoral (antibody) and cell-mediated immune responses against selected antigens. This method has shown limited potential as a SAV and in most cases, requires a booster consisting of either the antigen or additional DNA;
  • Controlled release vaccine to release organism antigens at selected times: The controlled release vaccine technology has shown the greatest promise. Several different methods have been used to achieve the controlled release of vaccine antigens including liposomes, unilamellar vesicles, emulsions, and polymers have all been tested as single immunization vehicles. Due to the instability of liposomes, lipid vesicles and emulsions in the body, these systems only produce an initial exposure to the antigen and usually require enhanced immunization in order to achieve protection of the disease. Polymer systems for vaccine delivery have been studied for more than 20 years and have shown promise as SAVs.
