Controlled Release Vaccines Development

Creative Biolabs gives a high priority to develop vaccines both in terms of its importance for public health and as a service to the biopharmaceutical industry. Our scientists are experts in their field and maintain their expertise through relevant research projects and collaborations. Creative Biolabs is committed to offering a consistently rapid, high-quality and cost-effective service with a focus on the development of controlled release technology of vaccines.

Vaccination has led to a substantial decline in the incidence of many infectious diseases in the past few decades, but successful vaccination often requires multiple immunizations, i.e. primary immunization and boosters to provide complete protection. Therefore, the development of new vaccines, particularly the conversion of multi-dose vaccine to single-dose vaccine has become an area of research. The development of injectable, sustained-release and controlled-release vaccines can be considered as the most promising strategy in vaccine delivery. By gradually releasing antigens, these systems reduce the frequency of injections, improve patient compliance, increase efficiency due to long-term antigen exposure, reduce adverse reactions and save costs.

Controlled Release Vaccines - Creative Biolabs

Characteristics of Controlled Release Vaccines

The attributes of this "perfect" vaccine technology include:

  • Consisting of a vaccine that provides complete and long-term protection after a single immunization;
  • Stability at high temperatures to reduce or eliminate the need for refrigeration;
  • Low production costs to allow widespread use;
  • Universal use of this technology for most existing vaccines.

Single Administration Vaccine (SAV)

Researchers have studied the possibility of developing single-dose vaccine (SAV). The main methods include:

  • Viral vectors encoding all or part of the pathogen genome;
  • DNA vectors carrying genes of pathogen: DNA vectors for immunization have recently attracted a great deal of attention as they are capable of producing humoral (antibody) and cell-mediated immune responses against selected antigens. This method has shown limited potential as a SAV and in most cases, requires a booster consisting of either the antigen or additional DNA;
  • Controlled release vaccine to release organism antigens at selected times: The controlled release vaccine technology has shown the greatest promise. Several different methods have been used to achieve the controlled release of vaccine antigens including liposomes, unilamellar vesicles, emulsions, and polymers have all been tested as single immunization vehicles. Due to the instability of liposomes, lipid vesicles and emulsions in the body, these systems only produce an initial exposure to the antigen and usually require enhanced immunization in order to achieve protection of the disease. Polymer systems for vaccine delivery have been studied for more than 20 years and have shown promise as SAVs.

ProSphere™ Printing Based Polymer Microsphere Platform for Sustained-Release Vaccine

Conventional vaccines often require repeated injections over long periods of time. A single-dose sustained-release vaccine that can achieve or exceed multiple injections in a single injection is one of the research hotspots in the field of vaccines. Creative Biolabs has developed a sustained-release platform, ProSphere™, using printing technology to produce millions of microspheres per second on a limited infrastructure with easy-to-remove environmentally friendly solvents. ProSphere™ has proven to be superior in extending the duration of vaccine release, enhancing vaccine efficacy, and having excellent commercial production characteristics.

Creative Biolabs carries out contract development for controlled release vaccines and have the expertise to investigate problems with research as they arise.
