Replacement of dC with 5-Methyl-dC

Introduction of 5-Methyl-dC

5-Methyl-dC is a methylated form of the DNA base dC. It is one of the important epigenetic modifications in living organisms, involved in the regulation of gene transcription. Oligonucleotides modified with 5-Methyl-dC have been used as effective research tools to investigate the effects of DNA methylation in the progression of some diseases such as tumors. Furthermore, 5-Methyl-dC modification is also applied in the synthesis of therapeutic antisense oligonucleotide. In vivo study has revealed anti-sense oligonucleotides containing a CpG motif can induce pro-inflammatory responses via activation of Toll-like receptor 9 (TLR9). While the replacement of dC with 5-Methyl-dC can prevent or dramatically reduce these unwanted immune responses. Besides, dC replaced with 5-Methyl-dC also increases Tm by as much as 0.5 °C per insertion, thereby increasing the stability of duplex oligonucleotide.
