Background

The angiotensin-converting enzyme (ACE) is a zinc-dependent peptidase of 90 to 200 kDa that adsorbs to the outer leaflet of the endothelial plasma membrane and then is shed in a soluble form into the circulation. ACE is also a bradykinin inactivator and an enkephalinase. This places ACE at the junction of regulation of vascular tone, hemostatic balance, and nociception. ACE is concentrated in capillaries of the lung, proximal tubules of the kidney, and circumventricular organs in the brain, but its mRNA can also be induced in macrophages in sterile inflammatory settings. As such, it has been suggested to have broader roles. Different mutations of the ACE gene particularly the insertion/deletion variant impact endurance abilities as well as the development of diabetic nephropathy and responses to SARS-CoV-2 infection. Pathologies such as myocardial fibrosis and cerebrovascular remodeling occur due to ACE overactivity while ACE down-regulation leads to refractory hypotension in septic patients. ACE inhibitors and newer dual ACE-NEP inhibitors thus remain mainstays of treatment and PET tracers and ACE-targeted protein chimeras are being explored for their potential to image pulmonary endothelial damage or deliver payloads to activated macrophages.

ACE Protein Function: A Central Node in Cardiovascular and Metabolic Control

ACE performs multiple functions beyond blood pressure control and researchers still lack full knowledge of its molecular basis:

  • Angiotensin-Dependent Neuronal Plasticity: ACE regulates Ang I-Ang II conversion at key brain sites within the angiotensinergic system to synchronize synaptic plasticity with stress response and cognitive processing.
  • Immunometabolic Regulation: Macrophages and dendritic cells which express ACE generate Ang II directly at the site of tissue inflammation while simultaneously managing chemokine secretion and directing the differentiation of T-cells.
  • Endothelial Oxidative Stress: Membrane-bound ACE has recently been proposed to serve as an anchoring protein for NADPH oxidase subunits, which in turn may control mitochondrial ROS efflux, thus determining endothelial cell fate (survival/death). ACE and mitochondrial quality-control machinery interactions constitute an intensely studied field with implications for cardio-renal protection.

ACE Stable Cell Line Product

Engineering physiologically relevant cellular systems is pivotal for advancing ACE-centered cardiovascular and neuro-immunological studies. Creative Biolabs therefore delivers bespoke, high-expression ACE stable cell lines—generated through our proprietary MemDX™ Cell-Line Engineering Suite—that preserve native membrane topology and full enzymatic activity. The stable cell lines we offer offer assurance of uniform ACE density, correct sub-cellular localization and a low level of background activity. This allows for a reproducible and scalable tool for the interrogation of ACE driven RAAS signaling, profiling of anti-hypertensive antibodies and to support the development of new generations of antihypertensive drugs, speeding up your discovery process from target validation to lead optimization.

Frequently Asked Questions (FAQ)

  1. Why is ACE a high-priority research target?

A1: ACE is a major control point for blood-pressure homeostasis and inflammatory pathways that cause fibrosis. It is a validated drug target for treatment of hypertension and heart failure. Genetic variants such as the I/D variant are used to predict cardiovascular risk and drug response.

  2. Q2: What is the main activity of ACE?

A2: ACE degrades angiotensin I into angiotensin II and degrades the vasodilator bradykinin. Ang II and BK degradation together result in hypertension and vascular changes.

  3. Q3: Are Creative Biolabs’s ACE proteins/antibodies approved for clinical use?

A3: No—every ACE reagent, cell line, or assay service is strictly “Research Use Only” and is not intended for diagnosis, prevention, treatment, or cure of any human disease.

  4. Q4: Can Creative Biolabs generate custom ACE stable cell lines or assay cascades?

A4: Yes, all services can be customized to your specific requirements.
