Chlamydiae trachomatis is a Gram-negative bacterium that scavenges host metabolic products for its replication. It can only replicate in human cells and is related to a wide variety of diseases, such as sexually transmitted diseases and blinding trachoma. Different strains can cause different type of diseases, most in the genital tract, the eye or lymph nodes. The life cycle is made up of two types, one is the elementary body (EB) which allows it survive outside of the host cells, and another is the active reticulate body (RB) which is responsible for binding and entering the host cells. The Chlamydiae trachomatis Vaccines

Chlamydiae trachomatis (Ct) infection is the cause of sexually transmitted diseases, leading to epididymitis and orchitis in males, as well as pelvic inflammatory disease, infertility, and ectopic pregnancy in females. In this condition, the Chlamydiae trachomatis vaccines have been a global health concern for preventing these diseases. A number of Ct vaccines are developed based on the understanding of the Ct life cycle and the host immune response to infection. Recent studies have been focused on the pathway of immune responses as well as the relationship between Ct and its infections.

Many vaccines have been developed but there are still no available Chlamydiae trachomatis vaccines approved to be safe and highly effective in clinic. The first generation of Ct vaccine is based on the live or formalin-fixed whole bacteria but the results show that it only can present a short-lived protection in human trials. Besides, research also reveals that a genital chlamydia vaccine would ideally target for Chlamydiae trachomatis infection prevention. Up to now, a number of novel vaccine candidates, including MOMP-VD4 neutralizing antibodies and live attenuated (plasmid-deficient) trachoma vaccine, are underway, and they should be entering clinical evaluation in the coming years.
