Cancer immunotherapy is emerging in recent years and is thought to be a safe and effective way to overcome cancer. This idea sees that cancer has undergone many changes compared to the self components in the body, and the immune surveillance function of the body's immune system can naturally recognize these differences and thus clear cancer cells. Studies in immunocompromised and immunocompetent mice have confirmed the validity of this theory. Therefore, as one of the hopes of finally conquering cancer, immunotherapy has become a research hotspot in this field. Among the strategies are vaccination, oncolytic viruses, and adoptive cell transfer, all of which aim to stimulate the body's anti-tumor immunity. Developing cancer vaccine is what we are professional at.

An effective cancer vaccine relies on the selection of antigens that are recognized by high-affinity T cells. Such antigens include neo-antigen due to mutations, post-translationally modified epitopes associated with cancer, cancer-associated antigens, viral antigens, and the like. Among various types of cancer vaccines, peptide vaccines capable of encoding tumor epitopes have shown advantages in animal experiments, but their effectiveness in clinical trials is very limited. Nucleic acid vaccines have been a popular vaccine type for some time. Because nucleic acid vaccines are fast, simple, and inexpensive to prepare. Nucleic acid vaccines do not cause immune responses to vectors itself, and DNA / RNA also has an inherent adjuvant effect so that it is possible to recruit immune cells or even activate immune cells at the site of inoculation.

VaxBody™

The design of VaxBody™ is based on the anti-tumor effects of T cell epitopes and the unparalleled advantages of DNA vaccines. VaxBody™ is a DNA that encodes a human antibody, and in the gene portion of the CDR region of the antibody it encodes, we replace it with T cell epitopes genes that induce anti-tumor responses. The epitopes we employed include both MHC-I binding epitope capable of inducing CTL responses and MHC-II binding epitope capable of inducing Th cell responses. Whereas the coding sequence of the Fc region of the antibody that VaxBody™ will encode was modified to specifically bind to CD64 (an Fc receptor with high Fc affinity) expressed on the surface of activated APC. Therefore, VaxBody™ not only stimulates both CD4 and CD8 T cell responses, but also binds to antigen-presenting cells such as DC with high affinity, thereby enhancing antigen presentation.

More importantly, VaxBody™ can present epitopes to T cells in both direct and cross-presentation. It turns out that the more pathways that present the same epitope, the stronger the T cell response. For VaxBody™, when the DNA form is injected into APC by electroporation, etc., it can be expressed and processed intracellularly, and then directly presented to T cells by the APC. When it is injected into muscle cells, it can also express the protein form of VaxBody™ in these cells, which can be further secreted and then presented by APC to T cells. Experimental results show that the presentation of the same epitope using multiple pathways significantly enhances the T cell response against tumor epitopes.
