Introduction to Adeno-associated Virus Vector

AAV, a member of the parvoviridae family, is a non-enveloped DNA virus that plays an important role in mediating the process of infections. Previous studies have indicated that chromosomal integration of AAVs has been widely used for generating a variety of transduction systems in gene therapy. Meanwhile, recent studies have also revealed that recombinant AAVs have higher transduction frequencies and can cause a minimal immune response in various types of cells. In particular, the transferred gene can keep stable expression in human cells. As a result, a number of AAV serotypes, including AAV-1, AAV-3, and AAV-6, have been identified and cloned as vectors, thereby overcoming resistance to anti-AAV-2 antibodies. Additionally, the application of AAV in gene therapy creates a demand for high titers and purified AAV stocks. Many advanced technologies have been developed for improving the purification of AAVs. As noted, a packaging plasmid that contains all the requirements for AAV amplification and packaging has been generated. The data suggest that this plasmid is effective in producing high titer adenovirus-free AAVs.

In the past few decades, a number of gene transfer strategies have been developed basing on AAV vectors. AAV vectors have shown its potential in transducing dividing and non-dividing cells, triggering a long-term gene expression in humans. In parallel, much effort has been made to determine the purification of AAV vectors because of the complexity of vector production procedure. Currently, many state-of-art methods for AAV vector purification have been generated by our labs. In general, purification of AAV vectors by Gradients Centrifugation Assays and Affinity Chromatographic Assays can shorten the turnaround time and lead to the reproducible concentration. For example, a study for serotype independent production of AAV vectors by using HEK293 cell and iodixanol gradient purification has been conducted by our company. In addition, we also have successfully purified a variety of cystic fibrosis-based AAV vectors by using hydrophobic interaction chromatography. Basing on these assays, we are capable to provide a high quality of laboratory-grade AAV vectors in a very short time.
