Single domain antibody (sdAb) represents the smallest antibody fragment that retains high specificity and affinity. They were first identified in camelids as a novel antibody isotype with heavy chain only. sdAbs combine the advantages of conventional antibodies with small molecule drugs. Given to the smaller size (12~15 kDa), sdAbs are able to bind conformational epitopes that are not accessible to larger molecules, such as full-length antibodies (150 kDa) or Fabs (50 kDa). Moreover, they also have better tissue penetration abilities (even through BBB) and are much easier to manufacture. On the other hand, comparing to small molecules, sdAbs retains good specificity, affinity but minimum cytotoxicity as antibodies.

Native premade single domain antibody libraries are constructed from non-immunized camels/llamas. VHH gene pool will be isolated and amplified in vitro, packaged into phage vectors, creating a comprehensive VHH library. Of note, we also have 2 well-established human sdAb libraries that allow rapid discovery of large numbers of high-potency camelized human sdAbs. The HuSdL libraries are generated based on camelized human VH3 in FR2. Diversity was introduced by random mutagenesis in different CDR regions. Meanwhile, human HCDR3 was elongated and sequence-randomized. Therefore, the libraries can raise camelized human antibodies with total human origin, which guarantees low immunogenic potential in humans, especially for long-term and multiple-dose administration.
