Cysteine amino acid residues are not present in the Tico-C9 fragment, and the distribution frequencies of the other 19 amino acids range from 3.1% to 8.2%, which agrees with the theoretical distribution probability (1/19 = 5.3%).

Screening in Action: Case Studies and Results

Discovery of Binding Peptides for Two Similar Target Proteins
Creative Biolabs has developed the experience and knowledge to identify highly specific binding peptides for even the most challenging targets, including when two very similar proteins are involved. Our client was interested in discovering peptide binders that could be specific to two similar target proteins, T-A and T-B, in one go, a necessary step for their therapeutic development.
Screening Strategy An intensive 4-round alternative screening was carried out with the Trico-9 Peptide Library with biopanning performed alternatively between target T1-A and T1-B. This approach was designed to increase the odds of finding peptides with an affinity to both targets.

Throughout the screening process, we observed enriching effects against both target proteins. This was further corroborated by polyclonal phage ELISA.
2. Identification of Dual Binders
Through monoclonal phage ELISA, we successfully identified 40 out of 80 clones that are positively bound to both T-A and T-B. Subsequent DNA sequencing of these positive clones revealed 7 unique peptide sequences, highlighting the diversity of the identified binders.

3. Peptide Synthesis & Validation
The identified unique peptide sequences were then synthesized in both cyclic and linear forms, all achieving a desired purity of over 95%. We proceeded to validate their binding activities:

  • Peptide 1: Both the linear and cyclic forms demonstrated strong binding to targets T1-A and T1-B.
  • Peptide 2: Only the cyclic form exhibited effective binding.
  • Peptides 3-7: All five peptides successfully bound to both T1-A and T1-B, with their linear forms showing higher activities compared to their cyclic counterparts.
