Aeromonas salmonicida (A. salmonicida) is a gram-negative, nonmotile rod, and non-spore-forming that can grow as single or paired rods of varying lengths or occasionally as coccoid cells. It is the causative agent of furunculosis pathogenic that severely impacts salmonids and other species. The virulence factors of A. salmonicida allow colonization of water including surface water such as salt water, beaches, and freshwater wells. Furunculosis induces septicaemia followed in chronic cases with the characterization of boil-like inflammatory lesions. The symptoms of the disease include external and internal hemorrhaging, swelling of the vents and kidneys, ulcers, liquefaction, and gastroenteritis. Hence, the development of a vaccine is necessary which could reduce the incidence of furunculosis outbreak.

Live Vaccine

There is a live vaccine which was designed by a kanamycin-resistant aromatic-dependent mutant of A. salmonicida. Oncorhynchus mykiss was used to examine the nature and magnitude of immune responses in this vaccine strain. Administration of this vaccine in fish can be performed by co-habitation, immersion or injection. After vaccination, good proliferative and antibody responses were elicited and a clear vaccine dose effect was apparent. Most importantly, this live vaccine preferentially stimulated enhanced T-cell responsiveness, as evidenced by significantly great increases in T-cell proliferation.

Inactivated Vaccine

The inactivated vaccine was comprised of formalin-inactivated A. salmonicida as immunogen and with the oil adjuvant. It provides high levels of protection over longer periods of time. Alternatively, using rainbow trout as recipients, injection vaccines based on this vaccine were formulated with CpG oligodeoxynucleotides, the liposomal cationic adjuvant formulation 01 or with Freund’s incomplete adjuvant and administered intraperitoneally. Subsequently, individuals were examined for differences in Fulton’s condition factor, macro- and microscopic pathological changes, as well as protection against experimental infection with A. salmonicida. Reductions in microscopic and macroscopic adverse effects were seen for both the nucleotide- and liposome-based vaccine formulations. Furthermore, the induced protection appears similar to that of the benchmark formulation, thus introducing viable, potential alternative types of adjuvants for use in future fish vaccines.
