Screening in Action: Case Studies and Results

Case: Development of Neutralizing sdAbs Against a Viral Antigen
Objective To identify neutralizing sdAbs targeting a viral antigen, Viral Protein T1, using the Phage Display HuVHHL-S1 Humanized Synthetic sdAb Library. The goal was to develop high-affinity neutralizing sdAbs for potential use in therapeutic applications against viral infections.
Starting Materials Viral Protein T1, the key viral protein responsible for binding to the receptor on host cells, enabling viral entry.
In-Solution Screening Strategy Pre-selection steps were implemented to remove nonspecific binders before initiating the main screening process.
Positive screening was conducted with four rounds of biopanning.
Competition-based screening to identify sdAbs capable of inhibiting T1 binding to the receptor.

Following four rounds of screening, several sdAb clones were identified that bound specifically to the T1. Subsequent internal validation by the customer confirmed that one particular clone exhibited strong neutralizing activity. This clone effectively blocked the interaction between T1 and its receptor in cell-based assays, successfully preventing viral entry into host cells.
