Case Successes: Demonstrating Screening Effectiveness

Case: Library Screening Against Cell-Based Targets
Objective To identify high-affinity sdAbs targeting membrane proteins overexpressed on cell lines, utilizing the HuVHHL-S2 phage display library.
Starting Materials Target: Membrane proteins overexpressed on cell lines (target protein T1).
Control: Control protein and cell line background controls to account for non-specific binding.
Cell-based Screening Strategy Pre-counter Selection: To eliminate phages that bind non-specifically to the cell lines or the control proteins, ensuring the selected clones specifically target the overexpressed membrane protein.
Cell-Based Biopanning: Perform multiple rounds of biopanning to enrich for phages that bind to the overexpressed target membrane protein while minimizing binding to control protein and background cell lines.
Binder Validation: The enriched phages are tested using flow cytometry and cell-based ELISA to assess their binding affinity and specificity to the target protein on live cells.

After several rounds of biopanning and screening, multiple clones were identified with high binding affinity and specificity for the target membrane protein overexpressed in the cell lines. These clones demonstrated minimal binding to the control protein or background cell lines. The successful clones were further validated through phage ELISA and FACS, confirming their ability to specifically recognize and bind to the membrane protein on living cells.
