PI (Phosphoinositide) is a minor component accounting for 2-10% of the total membrane lipids. However, it plays a key role in several fundamental processes including signal transduction and membrane trafficking. PI consists of a glycerol backbone, a phosphate group substituted with an inositol ring, and two acyl chains at the sn-1 and sn-2 positions. The inositol headgroup can be phosphorylated by lipid kinases at positions D3, D4, and D5, which produces seven kinds of unique phosphoinositides (PIPns). Phosphoinositides are universal signal molecules in eukaryotic cells. For example, phosphoinositide can facilitate the assembly of distinct chemical platforms on the surface of the membrane, which can regulate the protein activity recruited from the cytoplasm spatially and temporally. Additionally, phosphoinositides also act as key allosteric regulators of enzymes and ion channels. Lysophosphatidylinositol (LPI) is an endogenous lysophospholipid and endocannabinoid neurotransmitter. It is believed to play a key role in metabolic functions and cell functions.
