Solids-stabilized Emulsion

Background of Solids-stabilized Emulsion

Emulsions are used in many different fields which are now well established and the solid particles of colloidal size may be employed to kinetically stabilize emulsions. Solid-stabilized emulsions are obtained by shearing a mixture of oil, water, and solid colloidal particles. The term ‘Picking emulsion’ has been widely used to describe solid-stabilized emulsion, a kind of emulsion stabilized only by solid particles locating at oil-water interface, and they may advantageously replace conventional emulsions containing organic surfactants. In Pickering emulsions, the stabilizing film in between the droplets comprises very rigid layers that provide a mechanical barrier against coalescence. The solid particles are irreversibly anchored at the oil-water interface and develop strong lateral interactions. Many solid particles can be used in Pickering emulsions including organic (e.g. polymer zein or starch) or inorganic particles (e.g. silica, titanium dioxide and clay particles). Indeed, the continuous phase of an emulsion stabilized by particles having a high degree of hydrophilicity will be the water phase, whereas hydrophobic particles will preferentially stabilize water drops dispersed in an oil phase.

Solids-stabilized Emulsion in Drug Delivery

Topical drug delivery is a challenging area with many advantages such as avoidance of first passage effect, stabilization of blood concentrations and attainment of local therapeutic effect with fewer side effects. Moreover, it also allows a reduced drug dose because of the shortened metabolization pathway of the transdermal route versus the gastrointestinal pathway, which increases the overall bioavailability of the drug. Despite all these advantages, topical drug delivery remains limited to few molecules, since skin acts as a barrier to the delivery of many therapeutic molecules. To overcome this obstacle, a favored strategy relies on selecting suitable vehicles for dermatologic therapy. Pharmaceutical emulsions emerge as a good solution for skin drug deposition in spite of their thermodynamic instability. Consequently, Pickering emulsions are regarded as an interesting strategy to incorporate drugs in pharmaceutical formulations. Substituting solid particles for traditional surfactants, they are more stable than coalescence and can obtain many useful properties. Besides, they are more biocompatible when solid particles employed are relatively safe in vivo.
